The Penningtonville Presbyterian Church was organized on May 25, 1851, by order of the New Castle Presbytery. A church was erected on an acre of ground purchased from Alexander Hanna and dedicated on September 15, 1852, and a burial ground was established contemporaneously on the land around the church.
The cemetery was expanded over the years to encompass more than four acres of land managed by the Presbyterian Cemetery Association under the name Penningtonville Cemetery.
In 1986, the the nonprofit Presbyterian Cemetery Association was given control of the neighboring Atglen Baptist Cemetery. Today, both grounds are managed as Penningtonville Cemetery, with interments in the Atglen Baptist Cemetery referred to the "Baptist Section" of Penningtonville.
Because the distinction between these two grounds is no more than a grass strip separating the two parcels, interments in the Penningtonville Cemetery may (also) be listed in Atglen Baptist Cemetery.
