He was the son of: Adam Alexander Mary Shelby Alexander.

Isaac's father Adam was a large land owner a Justice of the Peace and a member of the County Court, being known as "Justice Alexander" as early as 1755. In 1756 he was a Captain of a Company of Militia and took a prominent part in a conference concerning Indian Raids.
Adam Alexander was Lieutenant-Colonel of Minute Men in 1775 and Colonel in 1776.
