Brodie Duke was the second son born to George Washington Duke and Mary Caroline Clinton Duke. His mother died about a year after his birth. An older brother, Sydney Taylor Duke, lived less than ten years.

His third marriage was to Alice Webb, to whom he was later divorced. He then married Minnie Woodward. He is buried in the Washington Duke Mausoleum (2nd crypt from bottom on right) with Martha V. Duke who died in 1888.
