Daughter of Sir John Arundell of Lanherne, Cornwall, and his second wife, Catherine Grenville.
Mary was a maid of honor to Queen Jane Seymour before she became the third wife of Robert Radcliffe, Earl of Sussex, on 14 January 1537. She had two sons by Sussex, but one of them died young.
She married Henry Fitzalan, Earl of Arundel on 19 December 1545, as his second wife. There were no children born of this marriage.
Mary died at Arundel House, Strand. London, and was buried in St. Clement Danes, but her remains were later moved to the Fitzalan Chapel at Arundel.
