John was the son of John Compton Sr. He married Elinor McGuire around 1778, about the time that he served in Capt. James Moore's militia company. They spent the months of April through October every year from 1779 to 1782 at the forts on Cove Creek for protection from Indian raids during the Indian War. In 1781, John received by patent a 500 acre tract on Clearfork of Wolf Creek, Tazewell Co., Virginia. He also had 400 acres on the Bluestone River, and was appointed Constable of Wythe Co., land which is now in Tazewell Co., and was listed on the petitions to form Tazewell County from Wythe. John was a farmer and a slave owner and can be found in numerous early tax and land records, census, and jury lists. He built the Compton home for his growing family on the Bluestone River in 1803. In 1812, he patented 350 acres on the north side of Sandy River above the mouth of Blackberry Creek, an area now part of West Virginia. John Jr. died soon after writing his will on March 12, 1828, as it was proved in court the following June [Tazewell Co., Virginia Will Book I, p. 244]. His estate consisted of 9 horses, 1 bull, 30 sheep, 25 cows, 22 pigs, 68 geese, wagons, tools, 6 plows, a steel trap, mill stones, household items, a loom and 8 slaves. It is assumed by his descendants that he was buried on his farm in an unmarked grave, most likely next to his wife Elinor.

John and Elinor were the parents of Nancy md. Thomas Cartmill, (migrated to Cabell Co., WV); John md. Mary "Polly" Walls, (resided Falls Mills, VA); William md. Jane Virginia Allen, (old homeplace on the Bluestone, 3 miles west of Bluefield); Hickman md. Elizabeth Justice, (resided on Clear Fork of Wolf Creek near Bland County); Rebecca md. John Shannon, (Lawrence Co., KY and back to Tazewell Co., VA); Elinor md. Low Brown Jr., (Elinor died 1833, Tazewell Co., husband remarried and moved to Carter Co., KY); Hiram md. Janney Shannon and Margaret Emert McGranahan, (migrated to Lawrence Co., KY); Sarah md. Col. William Wilson Compton, (Carroll Co., MO); Elihu md. Rebecca Shannon, (Livingston Co., MO); and Joseph W. md. Nancy Shannon (Falls Mills, VA).
