William served as a sergeant and quartermaster in the War of 1812 with the 19th (Ambler's) Regiment of Virginia Militia, and with the 4th (Cocke's) Brigade of Virginia Militia. He later received a bounty of western land, which he sold.

He worked as a merchant in Richmond, owning several stores, and taking over his late father's business located at Broad (then "H") and First streets. He later lost the business thanks to an unscrupulous partner. In 1832, he and other former members of 1st Baptist Church founded "Old Sycamore Church", located on 11th Street between Broad and Marshall streets. The church building was used as a hospital during the Civil War.

William married Sarah Bryan in Henrico County on 26 October 1815. They had at least two children, Rachel and William B. After Sarah's death about 1831, he married Anna Maria Carlton. They had at least four children: James K., Sarah A., John C. and Mary V.

William died of "Dysentery", per Cemetery records. His grave in the plot he owned, next to his wife, and which he shares with his son James, was marked through the efforts of Bootwright descendant Frederick "Pete" Nuckols, Jr. and the "Friends of Shockoe Hill Cemetery."
