John was the son of Joseph B. May Sr. and Elizabeth Baker. He married 1st Mary "Polly" Farley in Wilkinson Co., MS on 22 February 1808. They were the parents of six children: Green Berry, Sarah Elizabeth, Joseph Monroe, Matilda, Louisa Ann, and John David "Dudley" May Jr. After Mary's death, John married Elizabeth Thomas Barnes on 20 May 1829 in Pike Co., MS. They were the parents of three children: James Madison, Everett Matthius, and Amanda A. May.

John D. May served in the 13th Regiment, Nixon's Company, of Mississippi Militia during the War of 1812. He fought in the Creek Wars and was at the Battle of New Orleans. Per a family diary, John was supposed to be the sharpshooter who unhorsed General Packingham, causing his death.

His grave may not be marked.
