Samuel Harter's 1st wife was Mary Antrim, born about 1811 in New Jersey. They were married on 30 Jun 1832 in Tippecanoe County, Indiana and was last listed with him in the 1860 census in Dallas, Huntington, Indiana. Their known children were Enoch Enos, Hannah Suphronia, Mary Ann, Phillip, Josephus, Esther, Alonzo, Joseph, and Samuel.

Samuel married 2nd Mary (Laymon) on 13 Jan 1865 in Wabash County, Indiana. They had no children, although Mary had children from her 1st marriage to Hiram Filson. In 1866 Samuel and Mary moved to Livingston County, Illinois.

Samuel died on 28 Jun 1870 at his home in Reading Township. His will, written 24 Jun 1870, lists his heirs as his wife Mary and his children Enoch, Hannah Young, Mary Ann Halsted, heirs of Phillip (deceased), Josephus, Esther Kiser, Alonzo, Joseph, and Samuel.
