John was born in Westchester County about 1660, the son of John Archer & Catherina Carle. His father was Lord of the manor of Fordham. John, the father, had mortgaged the manor to Cornelies Steenwyck and after John's death in 1684, Steenwyck became the owner by forfeit in 1685. Steenwyck's widow married again, and in 1694, conveyed Fordham to the Nether Dutch Reformed Church in New York, but conflicting claims had reduced the extent of the property. The church took over the manor, and the land where Fordham University is now situated passed completely out of Archer family control.

John was apprenticed to Hartman Wessels, a surgeon, on February 1, 1677. He married first, Margarita Steenwick. They had no children. She must have died about 1686, because in December of that year, he married Sarah Odell, daughter of William Odell. They had seven children. Sarah must have died before 1715, because in that year, John married Mary Fowler, daughter of Henry Fowler. They had one daughter, and John died before 1718, when his widow married Samuel Drake. John is thought to be buried with his wife Sara Odell in the family plot on Tetard's Hill, Spuyten Duyvil, Bronx County, located on what once comprised the extensive grounds of the family estate, Fordham Manor.
