James, a printer by trade, was the son of Humphrey Calder and Isobel Kerr. He married Isobel Arthur in Scotland in 1820. They had three children: Humphrey, William and John Arthur.

After Isobel's death in 1826, he married Mary Clarke in Campsie, Scotland on 17 June 1831. They had four children: Lillias, Jane, Elizabeth and Mary. After Mary's birth, the family emigrated to the United States.
He died from intestinal problems, per Cemetery records. His grave in the H. and William Calder plot, which he shares with Mary Calder, is unmarked.
