Sir Francis was born in Clapham when it was considered part of Surrey. He was the son of William Cook and Mary Ann Lainson.

He was a merchant who was known for his art collection. He married his first wife Emily Martha Lucas on August 1, 1841. His children were all by his first wife:

Sir Frederick Lucas Cook, 2nd Baronet (1844-1920)
Emily Jane Cook Sartorius (1849-1915)
Wyndam Francis Cook (1860-1905)

He married second Tennessee Celeste Claflin. They have two marriage records on two separate dates, the second time recognizing that she had previously been married to a Bortle.

In 1884 he founded the Alexandra House (now called Queen Alexandra House) named after the future Queen Alexandra who was then the Princess of Wales.

He died at his home Doughty House in Richmond, Surrey, England.
