Il se marie à Twickenham, en Angleterre, le 30 octobre 1899, avec sa cousine germaine, la princesse Isabelle d'Orléans.
Prince Jean Pierre Clément Marie d'Orléans, Duc de Guise, was the third son and youngest child of Prince Robert, Duc de Chartres, and his wife, Princess Françoise d'Orléans, daughter of François, Prince de Joinville, and Princess Francisca of Brazil. He was a grandson of Prince Ferdinand Philippe and great-grandson of Louis Philippe I, King of the French. Upon the death of his cousin and brother-in-law, Philippe, Duke d'Orléans, the Duc de Guise became, at least for his Orleanist and Unionist supporters, titular king of France as Jean III, although the title was disputed by members of the Spanish branch of the family, descended from Louis XIV.
He was married at Twickenham, England on October 30, 1899 to his first cousin, Princess Isabelle d'Orléans, with whom he had issue.
