Charles was a son of Clare Dunn and Charles Carroll, Sr. He emigrated from Ireland in 1715 for political reasons and settled in Annapolis. He was a surgeon, a merchant, and a land speculator. He was one of the five founding partners of the Baltimore Ironworks Company.

He married Dorothy Blake, and they had three children: Charles III, Mary Clare, and John Henry. After Dorothy died in 1734, Charles married Anne Plater.

Charles' will was witnessed on October 4, 1755, "in the presence and at the request of the same Testator", so he died sometime after that date.
