Philonzo Chamberlain was born in Onondaga County, NY, USA. At present, the identity of his parents is not known. Several of Philonzo's direct-male-line descendants have done DNA tests to try to get more information about his ancestors. Based on the results of those tests, it is highly probable that Philonzo was a descendant of the immigrant Richard Chamberlin of Braintree, MA.

According to family tradition, Philonzo's father drowned in one of the Finger Lakes, and his body was never recovered. However, a gentleman who is shown by DNA to be a close relative of Philonzo researched historical newpapers for any mention of the drowning of a Chamberlain man in one of the Finger Lakes during the relevant time period. He was unable to find anything.

Philonzo's mother was still living, and, again according to family tradition, was either a Hunt or married a Hunt after Philonzo's father's death. We have been unable to determine whether or not this is true. Philonzo was raised either by Hunt relatives of his mother, or by Alexander and Eunice Charlotte (Seeley) Wilson, the parents of Philonzo's future wife, Cynthia Wilson. We have also been unable to determine which, if either, of these traditions is true.

Some online family trees give the name Cobe, Colby, or Colbee for Philonzo's father. This appears to be based on a genealogical report done for Marvin Howard Chamberlain, Philonzo's youngest child, in 1901. The World Chamberlain Genealogical Society has reviewed that report and found that the connection was incorrect.

In that report, names are given for Philonzo's parents - Joseph Chamberlain and Susan Hunt. The source of those names is not known. It is believed that the person who prepared the report was unlikely to have found those names through his own research. It is possible that Marvin gave the names to that person, and that the source of them might have been Philonzo himself. However, at this time, that is conjecture. Furthermore, although a Joseph Chamberlain and a Susan Hunt of the right ages have been located, there is presently no evidence that they were ever in the same state.
