Married Mary "Molly" Miles June 22, 1771. They removed to Goshen, CT in 1774 and resided there for over 30 years.

Samuel Hopkins came to the frontier wilderness of Livingston Co, NY in 1811 from Litchfield Co., CT with his sons Samuel Miles Hopkins and Mark Hopkins, the first land agent of the Mount Morris land tract. Mark relinquished his land agency in 1817 and removed to Huron Co., Ohio. He died at Chillicothe, Ohio in 1831 at age 58. Samuel Miles Hopkins, an honored and well-respected attorney, died in Geneva, NY in 1837 at age 65.

Samuel Hopkins and Deacon Jesse Stanley, a Revolutionary War soldier from Lexington, MA, were great friends. Hopkins and Stanley Streets, which cross each other in Mt. Morris, were named after them.

Samuel married a Miss Pratt as his second wife and she survived him for over 20 years. He died in Mount Morris in 1818 and was the first one buried in the old cemetery.
