Eliza Gamble was the daughter of Joseph Gamble and Ann Hamilton and a sister to the Governor of Missouri, Hamilton Rowan Gamble. Born in Winchester, Virginia, she was the second wife of Samuel Baker and had five known children. They were: Ann Gamble Baker (Mrs. Lloyd Logan), James Carr Baker, Eliza Hamilton Baker (Mrs. David Kyle Pitman), Joseph Gamble Baker, and Mary Lisle Baker (Mrs. Benjamin A. Alderson).

In her father's will dated September 1828 he mentions her late husband Samuel Baker and it appeared he had not left her with the means to support herself or their children.
