Samuel Franklin Penfield was the son of Sturgis Penfield and Laura Giddings.
Samuel owned a general merchandise store where he sold clothing and items that he manufactured. He was a hatter, and was a manufacturer of woolen cloth and carpets.

He studied for the Congregational ministry, and was head of a boy's school, a high school principal and was engaged in real estate and the loan business. He belonged to the "Illinois Prohibition Colony," which was instrumental in making Kansas a prohibition state. Earlier, he helped make Vermont a prohibition state.

With son George, he formed the firm S.F. and G.F. Penfield Insurance and real estate business which was very successful and which he was active with until his demise.

On December 10, 1851 he married Lemira Briggs Clark. This union produced five children:
*George Franklin Penfield (1852–1911)
*Charles Seymour Penfield (1854–1912)
*Edward Marlin Penfield (1857–1910)
*Robert Penfield (1867–1876)
*Mabel Stewart Penfield (1869–1964)
