The son of William M. & Elizabeth (Parker) Henderson, in 1860 he was a farmer living with his family in North Middleton Township, Cumberland County, Pennsylvania. He stood 5' 7" tall and had black hair and gray eyes and apparently never married.

A Civil War veteran, he enlisted in Cumberland County April 21, 1861, and mustered into federal service at Harrisburg July 27, 1861, as a private with Co. A, 7th Pennsylvania Reserves (36th Pennsylvania Infantry), the company that his brother Robert commanded. He was promoted to corporal June 25, 1861, to 2nd lieutenant of Co. E on March 1, 1863, and to aide-de-camp on the staff of Brig. Gen. Samuel Wylie Crawford June 8, 1863. Gen. Crawford made special mention of Henderson in his battle report on Little Round Top at Gettysburg. Richard then was brevetted 1st lieutenant March 1, 1864, then captain, and major on March 13, 1865. He was granted a real promotion to 1st lieutenant March 1, 1864, but did not muster at the rank. He returned to the 7th Pennsylvania Reserves to receive his discharge, which occurred at term's end on June 16, 1864.
