On the 19 January 1843, John Eusebius Isenhour was born to Jonathan Eisenhower (1815 – 1894) and Margaret Whisenand (1818 – 1911) in Bloomington, Monroe County, Indiana. Before 1860, he relocated to Zionsville, Indiana where he became a well-known farmer. He lived in a hexagon-shaped house on 20 acres north of Zionsville (now Bailey Court) from where he managed his two farms north of old US 52. He was a masonic member of the Hosbrook Lodge of Free and Accepted Masons in New Augusta.

He had several children by many wives throughout his life. Their names are Margaret Isenhour (1866–____), Cora E Isenhour Powell (1868–____), Mary Catherine Isenhour Brunes (1870–1958), Luther Isenhour (1873-____), Daniel Isenhour (1874-____), Isabel Isenhour (1878-____), Omer Franklin Isenhour (1881-____), and Lula Isenhour Bailey (1885 – 1964).

On 16 June 1934, John Eusebius Isenhour passed away after an extended illness of one month. He was 91 years old. His funeral was held at the old log-structured Bethel M. E. Church of Zionsville. He was laid to rest in the church graveyard, Old Bethel Cemetery, which is now abandoned. The site is presently the knoll in the southeast corner of the front lawn of the Becker Place, south of Little Eagle Creek, on the west side of State Route 421 (Old Michigan Road), Eagle Twp., Boone Co., IN. The cemetery is on one of Augustine Davenport, Jr.'s original land entries and was the early family cemetery for the Davenports, Lowes, Brocks and Conrads. In 1888, Milton S. Davenport, sole surviving son of Augustine, tried to restore the cemetery, but was unsuccessful.
