Samuel was the son of Joseph Frost and Sarah Blakemore. In 1885 the family immigrated to the United States onboard the passenger ship Chester, arriving in New York City on 10 Jul. From there they moved to Louisville, Kentucky. On 29 Mar 1899 Samuel married Alvina Stein and they had three children: Leah, Samuel, and an unnamed infant boy who was stillborn. At the time of his death he was employed as a locksmith. Samuel died of tuberculosis at the age of 40.
