Nathaniel Wise Benton was one of four children born to Maecenas Benton and Elizabeth Wise Benton. He was the younger brother of the artist Thomas Hart Benton. He was married to Alma L. Caldwell Benton.

Nathaniel was a veteran of World War I, where he served as a Second Lieutenant in the Army. After the war, he practiced law in Springfield, Missouri. He was the prosecuting attorney for Greene County from 1934 to 1938. He was appointed assistant attorney general for the state of Missouri in 1944.
