Dorothy Yorke married Thomas Dudley on her 21st birthday, 25 April 1603, in Hardingstone, Northamptonshire, England.
They had five children:
Reverend Samuel Dudley (b. 1608)
poet Anne Dudley Bradstreet (b. 1611/1612)
Patience Dudley Denison (b. 1613/1614)
Sarah Dudley Keayne Pacy (b. 1620)
Mercy Dudley Woodbridge (b. 1621)

She died in Roxbury on 27 December 1643 of colic; "a godly Christian woman & left a religious savor behind her." She is believed to be buried in Eliot Burying Ground, however time and weather has erased the location of this grave.
