James is the son of James Agnew. He was born in Down, Northern Ireland. James was married twice. His first marriage was to Margaret Ochiltree. They had two children: John and Janet. His second marriage was to Rebecca Scott in 1737. They had 10 children: Samuel, Agnes, Martha, James Jr., David, Margaret, Rebecca, Sarah, Abraham, and Anne.

The image engraved above the inscription on his stone is the coat of arms of the Agnews of Lochnaw, Scotland. The history of the family is recorded in "The Book of the Agnews: James Agnew of Pennsylvania," by Mary V. Agnew. The coat of arms is also found in Burke's Peerage of Great Britain. Before James' family lived in Ireland, they were hereditary sheriffs of Galloway, Scotland, and James' ancesters were Andrew Agnew of Lochnaw and Mary Kennedy; Mary being a granddaughter of King Robert III of Scotland. King Robert III was the son of Robert II, who was a grandson of Robert I, more commonly known as Robert the Bruce. (Lineage information by Loretta Layman).
