Captain Samuel Jefferson sailed to the Leeward Islands in the West Indies aboard the ship "Hopewell" with his brother, John and neighbor, Thomas Warner, in 1624. He settled on Old Road at Middle Island. Owned the Red House Plantation and the Wingfield Estate at Middle Island, which is now Romney's Manor , St Kitts, West Indies. His two sons and wife joined him on St. Kitts. He is buried at St. Thomas Church, Old Road, St. Kitts, Leeward Islands.

Some believe Samuel may have been living in Antigua at the time of his death, others state he was living at the Red House Plantation at the time of his death. He is buried at St. Thomas Church, Middle Island, St. Kitts.

He reportedly had three sons. Only two are known for sure.
