In 1860, William Kelly ran for governor of New York State. He was the candidate of the faction of the Democratic Party, which supported Stephen A. Douglas for President. A separate Democratic Party faction ran James T. Brady for Governor, and supported John Breckinridge for President.

Kelly was active in the State Agricultural Society. He was also a charter member of the board of trustees that established Cornell University in 1865.

The People of the State of New York, represented in Senate and Assembly, do enact as follows:

Section 1. Mary R. Miller, Joshua S. Bowne, William Kelly, Freeborn Garrettson Sr., Lewis Livingston, N. W. H. Judson, Theophilus Gillender, Homer Gray, William R. Schell, Ambrose Wager, Henry M. Taylor, James A. A. Cowles, William B Platt, Theophilus Nelson M. D., John N. Cramer and Andrew J. Heermance, and their successors, are hereby constituted a body corporate by the name of The Starr Institute, to be located in the village of Rhinebeck in the county of Dutchess.

His mansion in Rhinebeck, Ellerslie, was purchased by future New York governor and U.S. vice-president Levi Parsons Morton.

William was under 19 years old when his father died and his eldest brother John was also a minor. His youngest brother, Robert, joined with the other 2 brothers in business in 1828.
