Son of Asa Pope and Sarah Mixter Pope.
Samuel moved to Burlington in 1833. He first worked as a clerk in the glass factory, then moved to Philo Doolittle's express office. His next venture was to own and keep a general store, which he gradually expanded to an express office. He then converted the business to an express office.

Samuel then became a cashier at the Merchants Bank, and was later appointed vice president. In addition, he was a trustee of the Burlington Savings Bank. He sold his express office about 1867 to accommodate his growing work in the Banking industry.

Samuel married Lucina Allen on April 13, 1842. They had 12 children, including George, Samuel, Sophia P (Mrs. Daniel Kern), Sarah Ann (Mrs. Carroll S Lewis), Mrs. William Marsh, and Matilda.
