Daniel Burke was born December 5, 1873 in New Berlin, NY, the son of James and Mary (York) Burke. He obtained his A.B. degree from Hamilton College in 1893, and his A.M. degree in 1894. He went on to obtain his LL.B. from NY Law School (1896) and his LL. D. from Hamilton in 1936.

He established a law practice, Burke & Burke in 1929 and served as President of Connecticut Mill, as director of the Summit (NJ) Trust Co., as a director and eventually Chairman of the RT French Company, a manufacturer of condiments, and of Atlantis Sales Company. He joined the Hamilton College's Board of Trustees in 1936, served as board chair from 1938 to 1946 and was elected as a life trustee in 1952. Hamilton's Burke Library was named in his honor.

Danial Burke was elected President of the American Bible Society on July 6, 1944. Burke, during his term, presented a set of bibles in 78 languages to President Eisenhower for the White House library in 1954, to which Eisenhower flipped to a random page and exclaimed to his wife, Mamie: "Look, Swahili!."
