Mary Jane Jackson was the only daughter of Mitchell Jackson and his wife, Virginia Angelica Whitehead. She had a twin brother, Burrell Jackson, and two younger brothers, Benjamin Franklin and Willis Barney. After her mother died in 1836, the family moved to Hind's County, Mississippi where her father married his deceased wife's older sister, Jane Whitehead Trotter. After Jane's death in about 1841 or 42, Mitchell Jackson and his four children moved back to Tennessee.

Mary Jane married Robert Newton Quarles Hunt on April 4, 1844 in Dickson County, TN at the age of 15. The couple had six sons who died as infants, as well as six daughters and one son who lived to adulthood.
