Samuel was the son of Jonathan Shaw and Mehitable Pratt, the grandson of Deacon Jonathan Shaw and Phoebe Watson, Benajah Pratt and Persis Dunham.

Samuel was the husband of Desire Southworth, the daughter of Ichabod Southworth and Esther Hodges. They were married in Middleboro on 21 April 1731 and had at least the following children:
* Samuel Shaw Jr 731-1778
* Ichabod Shaw 1734-1821
* Elijah Shaw 1736-1806
* Capt William Shaw 1738-1807
