He was the son of Calvin Barber and Polly Hall Barber.
Per the Sons of the American Revolution Membership application for Walter Matthew Dick, Amzi Barber married Nancy Irene Bailey on November 23, 1841.
They were the parents of three children.

Mr. Barber's father, the Rev. Amzi Doolittle Barber, was graduated from the Theological Department of Oberlin College in 1841. Oberlin was at that time celebrated for its advanced and fearless attitude on the slavery question, just then bitterly agitating all classes in the United States. The Rev. Mr. Barber, after leaving college, returned to Vermont, where for many years he was pastor of the Congregational Church at Saxton's River, Windham County. His wife was Nancy Irene Bailey of Westmoreland, Oneida County, New York, a descendant of English and French ancestors.

Rev. Barber Died at Oberlin Thursday Night
At 6 o'clock Thursday evening occurred the death of the Rev. A.D. Barber, one of the old pastors of the First Congregational Church of Lorain. Rev. Barber has been living with his daughter, Mrs. M.G. Dick, at 195 South Professor Street in Oberlin. A few days ago he was stricken with paralysis of the nerves of the brain. He was over ninety-one years of age and very feeble and after lingering till Thursday night he passed away.

The deceased was the father of Mrs. Maitland, of Lorain and also of Amzi L. Barber, the wealthy New York capitalist who is interested in the Barber Asphalt company. He has spent some of his time in Lorain with Mrs. Maitland, but has been making his residence with Mrs. Dick in Oberlin. The funeral services will be held at 2 o'clock Sunday from the residence in Oberlin.
