Rev Zephaniah M Humphrey was the son of Rev Dr Herman Humphrey, who was a long time President of Amherst College.

He graduated from Amherst College in 1845 and studied theology in Andover, MA. In the winter of 1849 or 1850 he went to Milwaukee to the Plymouth Church. In October 1853 he was ordained pastor of the Presbyterian Church in Racine, WI where he remained until the spring of 1856. He was then called to Milwaukee where he was installed as pastor of Plymouth Church and remained until spring of 1859. He then accepted a call to the First Presbyterian Church in Chicago, IL beginning his ministry as pastor elect on May 15, 1859 and was installed on June 7, 1859.

He moved to Philadelphia in 1868 where he was pastor of Calvary Presbyterian Church.

He passed away after a brief illness on November 13, 1881 in Cincinnati, OH. His funeral was in Chicago on November 17 and burial was in Graceland Cemetery.
