Freeman of the Company of Mercers of London. Churchwarden of St Mary-at-Hill, and later, upper churchwarden of St Dunstan-in-the-East. Usher of the King's Chamber, Teller of the Receipt of the Exchequer, Commissioner, Gauger, and Sewer. His properties included Abbots Inn, which was the former townhouse of the abbots of Waltham Abbey, located in the parish of St Mary-at-Hill in Billingsgate Ward, London; a tenement called The Julian in the parish of Dunstan-in-the-East in Tower Ward; a tenement in the parish of St Peter at Paul's Wharf in Baynard Castle Ward; Knotts Lands in the town and parish of Leyton, Essex; and a watermill on the Galway River in Ireland.

Chaloner died testate with a will recorded on June 24, 1550 [PROB 11/33/264]. The IPM of his London properties was taken at the Guildhall, February 14, 1551, before Andrew Judde, Mayor and Escheator. The will of his third wife, Isabell Smith Chaloner, was dated August 13, 1558 [PROB 11/41/95], and is a lengthy, descriptive testament, providing much detail on the extended Smith, Allen, and Chaloner families.

By his first wife Margaret, daughter of Richard Middleton of London, he was the father of three sons, Thomas, John, and Francis [who was the father of Dr Luke Chaloner (1550-1613) of Dublin], and two daughters, Elizabeth and Helen.

Roger Chaloner was the eldest son of Thomas Chaloner by his wife Agnes Thickness, sister of Ralph Thickness (d. 1545). He was a grandson of Rhys Chaloner of Denbigh by his wife Ursula, daughter of Richard Peake of Lleweni Green. He was a great-grandson of Dafydd Chaloner of Rhuddlan by his wife Rose ferch Ithel Anwyl, and a 2nd-great-grandson of Hywel Chaloner of Rhuddlan.
