James was the son of John "The Pioneer" Calfee and Sallie Riggan. He married Jane Davis.

James, his mother and siblings left the Shenandoah Valley and settled on the south side of the New River in Montgomery Co., Virginia, near Little Reed Island, now in Wythe County. He was the grantee of 100 acres from Feathergill Adams on September 23, 1785. On July 1, 1788, he obtained an additional 600 acres in the same area from Adams. James had a ferry that crossed the New River at the mouth of Reed Island Creek, where he provided a "Good boat and one hand," and was permitted to charge the same rates as the Ingles's Ferry in Montgomery County.

James was co-executor (with brother William Calfee) of his uncle William Calfee's will proven in 1801. In 1805, in Augusta Co., Virginia, James brought suit against his aunt-by-marriage Mary Calfee, who had remarried to George Carter, to recover land his Uncle William had willed to him and his brother William.

From Chalkleys: "James Calfee vs Carter - O.S. 97; N.S. 33 - Bill, 1805. Orator, James Calfee, is nephew of William Calfee, died 1801, very aged and without children. Orator and his brother William, now deceased, took care of him. He died April, 1801, testate, devising all his real estate to his wife, Mary. In 1805, aged 74, [she] married George Carter. There is also a bill by James Calfee and William Calfee, executors of William Calfee, Jr. deceased. William, Jr., deceased, was nephew of William Calfee, Sr. late of Wythe County, who died 1801. Mary Carter answers she had made her will devising her estate to said James and William, John Hurst of Kentucky, and a niece, Miss Mary Carson of Shenandoah. William Calfee's will, of Wythe County, dated 10 August, 1795."

In 1808, James was appointed a Justice of the Peace in Wythe County. He was reappointed in 1810, and in 1811 was appointed Commissioner of Peace in Wythe County, where he served until his death in 1820. His will is in Wythe County, Will Book 2, p. 334.

From a letter dated April 15, 1913, by Dr. John William Holmes to Joseph Starke Calfee of St. Louis, MO: "Our Great-Grand Father James Calfee and his wife Jane Davis lie buried on a tract of land which he bought from Feathergill Adams in 1795 about three yards from the bank of New-River on its South Side and about the same distance above and opposite the mouth of Pine-Run-Creek in Wythe County, VA."
