Many sources, including Mayflower applications, cite his birth date 12 December 1697, son of Richard and Hannah Cooper. Plymouth VR 1:14: "John born on the 12 of Dec. 1697 [of Richard Cooper & Hannah Cooper his wife [a later hand added "he d. 1724"]." John Cooper died Cambridge 13 March 1724, "in his 26th y." and his Last Will leaves his entire estate to wife Lydia, to be split among his brothers and sisters after her decease.

John Cooper was a son of Richard Cooper & Hannah Wood, who are buried on Burial Hill in Plymouth, Massachusetts.

He married Hannah Rider on 8 Dec 1737 in Plymouth, Massachusetts.

They had at least 5 sons: Capt. Richard Cooper, married Hannah Sampson; Nathaniel Cooper, married Margaret "Peggy" Glover; Pvt. John Cooper, married Sarah "Sally" Sampson; Thomas Cooper, married Experience Holmes; and Joseph Cooper.

Joseph was mentioned in a family bible, but no more is known about him. Perhaps he took to the sea. He had a grandnephew named Joseph who was a sea captain, and a great grandnephew, Joseph Calvin, who went to sea before becoming a minister. Rev. Joseph Calvin Cooper married Rachel Alexander Van Dyke. She was a daughter of Isaac Van Dyke and Elizabeth Alexander Cooper, who was born 25 Dec 1796 at Greenwich Plantation, Charleston, South Carolina. An old family tradition in that branch of the Cooper family states that they were cousins. Perhaps Joseph, son of John Cooper & Hannah Rider, moved to South Carolina and started a family there. Alexander may have been the maiden name of his wife, but no records have been found to support this theory.

John & Hannah may have also been the parents of Samuel Cooper, born about 1738, who married Mary Smith in 1774. She may have been the Mrs. Mary Cooper, wife of Maj. Samuel Cooper, who died in New York in 1811.

The descendants of Capt. Richard Cooper, who married Hannah Sampson and Pvt. John Cooper, who married Sarah Sampson, are descendants of Henry Sampson of the "Mayflower" and through him have a legitimate line of descent from the Coopers of Henlow, Bedfordshire, England.
