James was the son of Elizabeth and Dr. Dudley Humphrey. He had 2 sisters: Ann Eliza and Emily Almira.

He married Laura Easterbrooks on August 19, 1841 in Orwell Township, PA. They had 3 children: Ira Bronson, Charles Dudley and Cornelia Eliza.

James was a Colonel in the Pennsylvania Militia. He was a tanner and harness manufacturer.

He was one of several committee members on general Christian education through the Presbytery of Susquehanna who petitioned for a Christian literary institution. The charter was granted in 1850 and this became the Susquehanna Collegiate Institute.

1856 -1863 he was a ruling elder of the Towanda Presbyterian Church.
