He was born one of two children (he also had a brother Enoch Ilsley), to Isaac Ilsley Jr. and Abigail Moody Ilsley in Falmouth, Maine, (later the city of Portland, Cumberland County, Maine, and part of the State of Massachusetts). He was educated locally by receiving liberal schooling. After his education, he became involved in distiller pursuits and also in the shipping business. On October 6, 1762, he married Mary Jones in Falmouth, Maine, and they had two children together (Henry Ilsley was born in 1779, and William Ilsley was born after 1779). Entering politics, he served as a Member of the Committee of Correspondence and Safety, and then served in the military as a Major and mustering officer during the American Revolutionary War in Falmouth, Maine. He then served as a Delegate to the Massachusetts Convention that adopted the United States Federal Constitution in 1788. He also served as a Member of the Massachusetts State House of Representatives in 1793 and again in 1794. He then decided to run for a seat in the United States Congress and was elected. A Member of the Democratic Party, he then served Massachusetts's 15th District (Tenth Congress) in the United States House of Representatives from 1807 to 1809. He was an unsuccessful candidate for reelection to the Eleventh Congress in 1808. After his term in the United States Congress expired on March 3, 1809, he was succeeded in office by United States Representative Ezekiel Whitman. He then lived in retirement until his death. He passed away on May 10, 1813, at the age of 72, and was buried in the Eastern Cemetery in Portland, Maine. His wife Mary passed away on August 10, 1812, at the age of 71, and she is buried next to her husband.
