Mary was a teacher in Vermont. She accepted a mission in 1869, at the age of 35, from the Dutch Reformed Church in America to teach at the Hepburn Academy in Yokohama. She was the first single woman missionary in Japan.

In 1872, she left the Academy and formed her own school for women. Her efforts later led to the development of the Ferris Seminary (now known as Ferris University). Eventually, she left the school to join her husband in missionary work.
