Ephrem was the fifth of seven children born to Pierre Chapleau and Zoe Siguin in Lower Canada.

All of the sons were quite successful, one a government clerk and two were doctors including Ephrem. Another brother Joseph-Adolph Chapleau, a lawyer, became the 5th Premier of Québec.

In 1873 Ephrem married Rachel Marie Adelaide Leprohon in Ottawa.
