Daughter of Nicholas Chowne of Fairlawn, near Wrotham in Kent, Jane became the wife of Sir John Puckering on 21 Feb 1568 at Aldenham, Hertfordshire. Mother of five children, only two of whom were surviving at the time of her death.

According to probate records, she married after her hersband's death as her will is notated "Puckering, Dame Jane, widow of Sir John Puckering, knight, Lord Keeper of the Great Seal, and late wife to William Combe, esq., deceased."
