Amos was the son of Moses and Phebe (Ballard) Alderson. Moses Alderson dated his Will on July 29, 1825 (Hendricks County, Indiana) and names as his heirs: his wife Phebe, the oldest children, Mary M. and Aaron and the youngest children, Harriet and Amos. He also mentions his brother John. Moses states that daughter Mary M. is to have "linens and other items that belonged to her mother, my former wife." Being as Moses sets out oldest and youngest children, Phebe (Ballard), his wife when he died, is probably the mother of his two youngest children. Phebe (Ballard) Alderson was born 1795 and died 30 October 1857. Illustrated Historical Atlas of Hendricks County, Indiana, 1878, p.48.

Amos was the beloved husband of Elizabeth Jane (Shanafelt) Alderson who died January 7, 1857, three months after giving birth to their daughter Sarah Catherine Alderson. Baby Sarah Catherine died October 24, 1856. Elizabeth Jane Shanafelt and Amos married February 26, 1843 in Morgan County, Indiana had a son, Clarence Marion Alderson born 1855. Elizabeth Jane Shanafelt was the daughter of Henry and Hanna (Hartle) Shanafelt.

After the death of Elizabeth Jane, Amos married Sarah Jane Cowgill on February 9, 1858. Sarah Jane Cowgill was the daughter of Jonathan B. and Lucinda Cowgill. The children of Sarah Jane and Amos: Ella Catherine, Harvey B., Harriet (m. Charles Herbert Wilson), Mary L.(m. Edgar J. Andrew) and Dora Alderson.
