Isaac Knox was born in 1832, apparently in Potsdam Village, Oneida County, New York. During the Civil War - from 10/3/1861 to 9/1865 - he served in Company G, 3rd Minnesota Infantry, re-enlisting on 1/1/1864. He was discharged at Duvall's Bluff, Arkansas.

On December 8, 1865 in Wabasha County, Minnesota, Isaac Knox married Eunice (Boughton) Youngs, widow of Enoch Youngs, who was killed in the Civil War. Eunice had five daughters by Enoch Youngs: Clarisa Jane, Roxy Ann, Sarah Ellen, Mary Elizabeth, and Cora.

Together, Isaac and Eunice (Boughton Youngs) Knox had three sons:

> William Knox, born at Mazeppa, Wabasha County, Minnesota on November 23, 1866, and farmed in Otter Tail County, MN. William died May 13, 1937, Otter Tail County, Minnesota. (Married Elizabeth Marsh in 1902)
> Austin Charles Knox, born in 1868 in Minnesota, later farmed near Ray, North Dakota. Austin died January 4, 1941, Williams, ND. (Married Anna Marie Larson in 1891)
> Warren Knox, born in 1870 in Minnesota.

On June 19, 1876 in LaSalle County, Illinois, Isaac married his stepdaughter, Sarah Youngs. Together, Isaac and Sarah (Youngs) Knox had two daughters and one son:

> Viola Knox, born August 5, 1876 in Illinois (married LD Richmond in 1896, Tom Hinds and Tom Gore).
> Nina Knox, born December 16, 1878 in Illinois, died in 1962. (Married Chester Swartout in 1899)
> Ira Knox, born February 16, 1885 in Illinois, died January 13, 1890 in LaSalle County, IL.

Isaac died on February 20, 1912. According to his obituary, at the time of his death he had been living in Marseilles, Illinois for at least 25 years. He was employed as a flagman at the Rock Island crossing at Chicago Street in Marseilles.
