John B. Palmer came to Rochesterville (today's Rochester, New York) circa 1816 from Maryland. He was a tailor by profession, a skill he handed down to his son, John Fletcher Palmer. He also ran a mercantile business in early Rochester.

His marriage to Margaret Wallace took place in Prince George's County, Maryland, on 1 May 1802.
