William Painter, the seventh of the eight children of Susanna (née Moorman) and Jesse Painter, was born on 18 May 1856 in Walnut Township, Madison County, Iowa. His parents settled there in 1853, coming from Greene County, Ohio, followed in 1855 by his maternal grandparents. William's father died in 1860 in Madison County, and his mother died there, too, in 1865. William and the younger children in the family were looked after by their eldest siblings, Rhoda and John Thomas Painter (then adults).

William Painter was married three times—

• Mary A. Deardoff, 19 June 1880, Madison County, Iowa. (Mary Aphracia Deardorff was the daughter of Rebecca Jane (née Gorsuch) and William Pleasant Deardorff.) Mary and William Painter had two sons: Jesse Carl and Clair William. Mary died in 1911.

• Rebecca E. Simmons Cason 19 March 1913, Madison County, Iowa. (Rebecca Elnora (née Simmons) was the widow of William Thomas Cason.) It appears that William and Rebecca divorced, and that she retained Painter as her surname. She died in 1940.

• Carrie McCargar, 7 September 1921, Mahaska County, Iowa. (Carrie (née Brownell) was the widow of Charles W. McCargar.) Carrie survived her husband, and died in 1946.

William Painter practiced medicine in Hubbard, Hardin County, Iowa, until increasing deafness compelled him to turn to the druggist and jewelry businesses. He died at the age of 82 in Dubuque, Dubuque County, Iowa, and was buried with his first wife Mary.

"The settlement of Friends, or Quakers, in Madison County, Iowa, began in the year 1853, when Jesse Painter and wife settled about midway between the present locations of the towns of Peru and Barney, in the southeast part of the county […] developed into Oak Run Monthly Meeting […] The next persons to settle at Oak Run were Thomas and Dosha Moorman, in 1855." Jesse Painter was a Madison County, Iowa, entryman. In June 1856, he received four patents for Federal land—a total of 200 acres—in the county's Monroe and Walnut Townships in sections bordered on the south by Union and Clark Counties.
