Charles de Lorraine, Duc de Guise, was the son of Henri I de Lorraine, Duc de Guise, Prince de Joinville, and his wife, Catherine de Cleves, Comtesse d'Eu. He was married on January 6, 1611 to Henriette Catherine, Duchesse de Joyeuse, the widow of Henri de Bourbon, Duc de Montpensier, with whom he had issue.

He died while in exile from France, in Cuna, near Siena in Italy. He was originally interred in the Santi Giacomo e Cristoforo de Cuna, but his remains were returned to France in 1641 and he was interred in the ancestral burial ground of his family at the Collégiale Saint-Laurent in Joinville. This structure was completely destroyed during the French Revolution and his ashes and those of his other family members were interred in 1792 in the Cimetière Chapelle Sainte Anne.
