Thomas III was the oldest son of Thomas II and Mary Matthews Simpson. He born on St. Thomas Plantation.

He married Sarah Connors, daughter of Richard and Mary Jenkins Conner, granddaughter of Thomas Jenkins, Esq., of the Jamestown Colony, in 1720. From this marriage, he inherited part of "St. Thomas" plantation. Not much else is known about Thomas except that he sold off over 200 acres of "St. Thomas" in 1743.

Sarah died in 1744, having bore Thomas four sons and a daughter:
Andrew (b. 1720)
James (b. 1722)
Mary Ann (b. 1724)
Thomas IV (b. 1724)
Ignatius (b. 1729)

Thomas remarried in 1746 to Mary Fanning and had no children.

Thomas died in 1760 and his will, probated December 14, bequeathed land to his eldest sons, Andrew and James, and money to his remaining three children. His will was probated in 1761 and was witness by Daniel of St. Thomas Jenifer, Clerk of the Court, Maryland delegate to the Constitutional Convention and signer of the United States Constitution.

Sarah Simpson is listed as the granddaughter in the last will of Thomas Jenkins, probated 1726.
