Nancy was a daughter of Greenup and Tomasin (Day) Duncan. She married Major Spencer 20 February 1844 in Morgan County, Illinois. They had nine children: Nancy Jane, who died at 7 months old; Elizabeth T., wife of Louis Herring; Sarah Jane, married to Carroll Gilmore; Henry, husband of Virginia Barnett; an unnamed baby boy; Major Leander, who married Mary E. Moore; Greenup, husband of Laura B. Bruce; Maria Octavia, wife of Andrew B. Reid, and Philip Fry who married Julia Ann Story.
