He was the 3rd Earl of Sinclair. In 1489, he was granted by charter, a 13 year lease (renewed for 19 years in 1501) of Orkney and Shetland, the Justiciarship thereof and Kirkwall Castle. On 26 January 1488/89, he was decreed by Scots Parliament to be the heir of his father and grandfather and ‘Cheiff of that blude'. He held the office of Master of Artillery in March 1510/11. He gained the rank of Captain in 1512 in the service of the ship Great Michael.

Lord Henry Sinclair was the husband of Margaret Hepburne. He was the father of Agnes, Katherine, Jean, Helen and William (the 4th Earl of Sinclair). He was the son of William Sinclair, 2nd Lord Sinclair and Christine Leslie Sinclair.
