James Duke was the son of Lewis Duke, Sr. and Elizabeth Holloway, who came to Henderson County, Illinois from England in 1844. Here, the family acquired land and with it, wealth and prestige; however, they were also known as generous and giving. Family records indicate that at one point he gave 80 acres to a Taliaferro relative, one who was poor of sight. Another family record indicates that he and his wife gave up their rights to the Coghill family estate so that a Coghill family member, who was deaf and mute, would have a nice farm to live on and work.

On 6 Jan 1853, in Henderson County, James married Millicent Ellett Coghill of the prominent Benjamin Coghill family. They had two children: Mary Elizabeth and Alpheus Wellington. Mary married Kingsley Adolphus Pence, son of Robert Titus and Elizabeth Conger Pence from Henderson county; there were no children. Wellington married Iva Iseminger, daughter of James and Martha Tharpe Iseminger of Henderson County; they had one son, James W, who was just a toddler when his father died.

James and Millicent farmed for many years around Monmouth before retiring to live in town. His parents preceded him in death.
