Charles was the son of famous Kentucky surgeon, Dr. Benjamin W. Dudley and his wife, Anna Short Dudley. He went to medical school and practiced medicine until his marriage to Margaret Julia Johnson, daughter of Henry and Julia Flournoy Johnson at the Johnson Plantation on Lake Washington, Mississippi in 1855. Margaret had been previously married to James P. Erwin who died in 1854 and the couple had several young children. This fact did not seem to daunt Dr. Dudley. The couple had a child of their own, Charles Wilkins Dudley, Jr., in 1856. He also managed to build for his new wife the beautiful plantation home known as "Mount Holly", before the War. It is one of the few antebellum homes left in the region known as the Yazoo Mississippi Delta. Margaret died in Fayette County, Kentucky in 1863. About four years later, Charles married Margaret's sister, Louisa in Washington County, Mississippi. Louisa and Charles had no children of their own.
