A native of Longford, Ireland, Father Joseph Mullooly moved to Italy, where he joined the Order of Preachers and was ordained to the priesthood. Assigned to the Basilica of San Clemente by the Master General of his Order, he earned a lectorship in sacred theology from the convent of Santa Maria sopra Minerva in 1849.

Appointed superior of San Clemente the following year, Mullooly initiated excavations at the Basilica in 1857. Ten years later, he brought to light a Paleochristian Basilica, dating back to the times of Saint Clement himself. In 1863, the vault of Saint Cyril was also located. In 1873, out of appreciation to his outstanding works, his brethren asked the Master General to confer upon Father Mullooly the title of Master of Sacred Theology.

Renowned also for his writings, suffering from malaria and pleurisy, Mullooly fell to the ground shortly after celebrating mass. Confined to his cell, he passed away five days later at San Clemente on June 25, 1880. Originally buried at the Campo Verano Cemetery, in 1912, his remains were transferred to the Paleochristian Basilica which he brought back to light, and reburied in the east end of the south aisle from where they were afterwards moved to a vault in front of the high altar of the excavated church.
